The innuPREP Bacteria DNA Kit has been designed for isolation of DNA from Gram-positive and Gram-negative bacteria cells derived from liquid cultures. The extraction procedure starts with centrifugation of bacterial cultures and a subsequent pre-lysis step. Especially when processing Gram-positive bacteria (e.g. Staphylococcus strains) this step significantly en-hances the yield of bacterial DNA. The pre-lysis step is followed by Pro-teinase K digestion and subsequent DNA isolation using spin column technology. By this means the innuPREP Bacteria DNA Kit allows a fast and effi-cient isolation of DNA from a broad spectrum of bacteria. The bacterial DNA is available in approximately 15 minutes after lysis of starting ma-terial. The isolated DNA is suitable for all downstream applications commonly used.

Using the innuPREP Micro RNA Kit allows researchers to isolate small RNA molecules and achieve high yields. The new, optimized binding buffer system makes it possible to achieve high rates of recovery for small RNA molecules such as mRNA, tRNA, rRNA and snRNA. The first step utilizes well-established spin-filter-column-technology to selectively remove genomic DNA; the RNA is then bound, washed and finally removed from the filter membrane using RNase-free water. Users have the flexibility to adjust the elution volume within a range of 30 µl to 80 µl. Also, the extraction chemistry (DC-Technologie) renders the use of highly toxic beta-mercaptoethanol unnecessary for isolating RNA. The innuPREP RNA Mini Kit 2.0 is the all-rounder for the extraction of total RNA. The specially optimized lysis buffer system guarantees the isolation of intact RNA and sustainably inactivates endogenous and exogenous RNases. Genomic DNA is removed via an optimized precolumn included in the kit. Accordingly, the otherwise necessary DNase I digestion can be completely omitted. Subsequently, the RNA is bound to a second Spin Filter membrane, washed and finally eluted in 30 - 80 μl RNase-free water. Thus, depending on the starting material, the extraction process can be completed in as little as 15 - 40 minutes. Both the protocol procedures and the Spin Filter columns have been optimized compared to the previous version and guarantee improved quality and higher quantity of isolated nucleic acid.

The innuPREP DNase I Digest Kit is a highly efficient tool for removing DNA from RNA samples that have been contaminated with DNA. Enzymatic DNA digestion can be conveniently integrated into the RNA isolation process. The DNase I is diluted in the buffer provided and then applied directly onto the spin filter column containing the bound nucleic acids. Digestion is followed by additional wash steps before highly pure RNA is eluted. The reagents provided are free of RNase and therefore also contribute to the quality of the RNA. Efficient DNA removal makes the innuPREP DNase I Digest Kit suitable for applications in which even the tiniest amounts of DNA could distort the results. Potential applications include preparing DNA-free RNA, DNase footprinting, Nick translation or degradation of DNA template when transcribing to cDNA.

Circulating cell-free DNA is of special interest as a diagnostic target, but the amount of free-circulating DNA is usually very low and varies among different individuals. Furthermore, these nucleic acids are present as short fragments, typically smaller than 1000 nt, making the efficient extraction process challenging. Because of the high sample volumes, the protocols of commercially available kits are very labor-intensive as well as time consuming and need a lot of reagents. The PME free-circulating DNA Extraction Kit is based on a new, patent-pending technology called PME – Polymer Mediated Enrichment. As first step cell-free DNA in the entire sample is captured by a polymer. Afterwards this complex is collected as a pellet by centrifugation. Subsequently the captured nucleic acid is dissolved in a special buffer thus reducing the sample volume in the following extraction significantly.

When it comes to food analysis, reliable and highly efficient DNA extraction from respective food samples is the foundation of the generation of valid results. The PME Food DNA Extraction Kit has been specifically developed for the simple and rapid isolation of DNA from liquid as well as solid food samples such as milk, milk powder, juices, wine, various flours, meat, chocolate, bread, and many more. It is based on the innovative technology called PME - Polymer Mediated Enrichment. Free DNA from liquid samples is captured by a polymer after protein digestion. Both the polymer-DNA complex and other intact cells are then pelleted by centrifugation and dissolved in lysis buffer. This workflow guarantees maximum yield of cell-free DNA frequently present in processed food products. In contrast, solid food samples are directly dissolved in lysis buffer before DNA is purified based on spin filters. Thanks to the optimized chemistry of lysis and binding buffer, critical food samples such as chocolate and tomato paste can be processed easily and the presence of inhibitory substances within the eluate is minimized. Thus, DNA purification by the PME Food DNA Extraction Kit offers ideal conditions for the successful implementation of downstream applications such as PCR-based food authenticity testing.

The PME Exosome Enrichment Kit is based on a new technology, called PME – Polymer Mediated Enrichment. Addition of PME to any type of biological fluid (e.g. serum, plasma, urine, ascites, cell culture supernatant, fractions from EVs size separation by size exclusion chromatog-raphy (SEC)), efficiently immobilizes exosomes within the net created by a special kind of polymer. Subsequently, the sample is centrifuged to form a pellet containing PME/exosomes complex. The obtained supernatant contains free proteins of the sample, including microRNA-carrying proteins, like Ago2, HDL or LDL. The exosomes-containing pellet can be resuspended in a dissolving buffer and used directly for downstream applications like microscopy, NanoSight, Western Blot, etc. The PME/exosomes pellet can be also lysed and exosomal microRNAs can be isolated with the use of innuPREP cell-free microRNA Kit. MicroRNAs from the exosome-depleted supernatant, as well as from the starting material can be isolated by the innuPREP cell-free microRNA Kit as well.

The innuPREP Sewage Water DNA/RNA Kit has been designed for the isolation of DNA and RNA from sewage water samples. /
The kit allows the isolation of DNA/RNA from viruses, bacteriophages and free-circulating DNA/RNA (e.g. plasmid associated DNA coding for bacterial resistances) from sewage water sediment as well as from the cleared supernatant after spinning down the sediment. The latest data shows that nucleic acids from this source could be detected in the sed-iment and the supernatant from sewage water samples.
The kit contains 3 protocols. The first protocol is for the isolation of nu-cleic acids from sewage sediment, the second from the corresponding supernatant and the third protocol allows the simultaneously isolation of nucleic acids from sewage sediment and from the corresponding su-pernatant.
The kit combines a unique patented technology for enrichment of bio-molecules with a very efficient extraction of DNA and RNA using mag-netic particles.
The extraction process starts with 20 ml of sewage water sample. After sedimentation of solid material 10 ml of the resulting supernatant and the resulting sediment (appr. 100 – 200 mg) will be used for the ex-traction of DNA and RNA.
Depending on the respective protocol the supernatant and the sedi-ment alone or both combined could be used for isolating nucleic acids.
